Study on the Optimization of Fermentation Conditions for
Penicillium citrinum Producing Nuclease P1

Abstract£ºThe fermentation conditions for Penicillium citrinum ZM-48 to produce nuclease P1 that is a mutant strain selected by means of UV radiation are studied and the optimum factors are selected through mono factorial test and orthogonal design direct. The optimal medium for producing nuclease P1 is composed of glucose 3%, bran infusion 3%, peptone 0 3%, yeast extract paste 0 3%, K2HPO4 0 05%, KH2PO4 0 05%, CaCl2 0 08%, MgSO4 0 04%, MnSO4 0 04%, and ZnSO4 0 02%; the other optimum conditions are initial medium pH 5 69, inoculum 10%, culture temperature 34 degrees centigrade, rotational speed of shaker 180 r/min, and culture period 72 h. Under the above optimum fermentation conditions the enzyme activity of nuclease P1 form Penicillium citrinum ZM-48 strain can be enhanced to 240 U/ml from 140 U/ml.
